Determining Solanesolin in Tobacco Leaves and Its Extracts Using HPLC Standardized Method

Abstract
We elucidate a Chinese standard: Determining solanesolin in tobacco leaves and its extracts using HPLC, so that the users can understand and use this standard more proficiently. The best extraction method and condition of saponification was screen out. The HPLC conditions were as follows: Waters Atlantis T3 C18 column (4.6 mm i.d.×150 mm,3 μm ) was used at 35 ℃ with isopropanol-methanol (30:70, v/v) as the mobile phase with a flow rate of 1.0 mL/min, detector UV-213 nm. The detection limit for tobacco leaves and its extracts was 1 mg/kg. The linear range was from 0.1 to 5.0 g/100g, and correlation coefficients were greater than 0.9999. The recoveries were greater than 95% with relative standard deviations of less than 5%. The method is rapid, simple and accurate.
