      侵染青岛烟草的主要病毒种类鉴定

      Identification of Main Virus Species in Qingdao Tobacco

      • 摘要: 为明确为害青岛烟草的主要病毒种类与分布,2023—2024年对3个主要烟叶种植站开展病情普查,利用烟草病叶宏病毒组和宏基因组测序与PCR检测相结合的方法,鉴定烟草病毒病的病原。结果显示,宏病毒组测序共检出6科6属9种植物病毒。其中,黄瓜花叶病毒属、烟草花叶病毒属和马铃薯Y病毒属为优势属,丰度分别为70.0260%、12.4185%和0.3639%。9种植物病毒依次为:黄瓜花叶病毒、烟草花叶病毒、番茄褐色皱果病毒、地黄花叶病毒,马铃薯Y病毒、辣椒脉斑驳病毒、番茄斑萎病毒、萝卜黄化病毒和番茄黄化曲叶病毒。宏基因组测序显示病毒丰度为0.3136%,其中,植物病毒为0.2861%,涉及6科17属39种。烟草上已报道的3种DNA病毒:烟草脉明病毒、番茄黄化曲叶病毒和紫云英矮缩病毒丰度较高,分别为0.1264%、0.0009%和0.0001%。大田普查和PCR检测显示,危害青岛烟草的优势种类为烟草花叶病毒、马铃薯Y病毒和黄瓜花叶病毒,次要种类包括首次在青岛烟草上检出的3种病毒:番茄斑萎病毒、辣椒脉斑驳病毒和紫云英矮缩病毒,以及番茄黄化曲叶病毒。

         

        Abstract: To clarify the main virus species and distribution of tobacco viral diseases in Qingdao, the disease surveys were conducted in three main tobacco-growing stations of Qingdao from 2023 to 2024. The pathogens causing tobacco virus diseases were identified using virome sequencing, metagenome sequencing and PCR detection. The results showed that, a total of 9 plant viruses from 6 families and 6 genera were detected through virome sequencing. Among them, Cucumovirus, Tobamovirus, and Potyvirus were the dominant genera, with abundances of 70.0260%, 12.4185%, and 0.3639%, respectively. The 9 plant viruses were cucumber mosaic virus (CMV), tobacco mosaic virus (TMV), tomato brown rugose fruit virus (ToBRFV), rehmannia mosaic virus (ReMV), potato virus Y (PVY), chilli veinal mottle virus (ChiVMV), tomato spotted wilt virus (TSWV), turnip yellows virus (TuYV), tomato yellow leaf curl virus (TYLCV). Metagenomic sequencing revealed a virus abundance of 0.3136%, with plant viruses accounting for 0.2861%, encompassing 39 species from 17 genera and 6 families. Among the DNA viruses previously reported in tobacco, tobacco vein clearing virus (TVCV), TYLCV and milk vetch dwarf virus (MDV) showed relatively high abundances, with 0.1264%, 0.0009%, and 0.0001%, respectively. Field surveys and PCR detections indicated that the dominant viruses in Qingdao tobacco were TMV, PVY, CMV. Secondary viruses included TSWV, ChiVMV, MDV, and TYLCV, with the first three being detected for the first time in Qingdao tobacco.
