    重庆丰都烟区田间烟草病毒病及其Dot-ELISA检测

    Detection of Field Viral Diseases on Tobacco in Fengdu County of Chongqing with the Dot-ELISA Technique

    • 摘要: 为明确重庆市丰都烟区田间烟草病毒病的发生情况,开展了病毒病调查,并于还苗期、伸根期、旺长期和成熟期共采集病毒病样品117份,利用TMV、CMV、PVY、TEV、PVX、TuMV和BBWV-2等7种病毒的特异性抗体对其进行Dot-ELISA检测。调查发现,病毒病症状主要包括花叶、畸形、坏死斑、脉坏死、矮化和蚀纹等类型。9个调查点烟草病毒病的发病率在28%~84%,病情指数最低为6.67,最高为60.89。Dot-ELISA检测结果表明,还苗期样品未检出病毒;伸根期没有检测到PVX;旺长期和成熟期均检测到7种病毒,其中TMV和CMV的检出率较高,其次为TuMV和PVY,PVX的检出率较低。此结果表明,烟草病毒病在丰都烟区发生较普遍,且TMV和CMV为田间烟草病毒病的优势毒源。

       

      Abstract: In order to survey the occurrence of tobacco viral diseases in Fengdu, Chongqing, 117 tobacco leaf samples with viral symptoms were collected at different growth stages (seeding, stem elongation, vigorous growing and mature) and detected with the Dot-ELISA technique using the specific antibodies against 7 viruses including TMV, CMV, PVY, TEV, PVX, TuMV and BBWV-2. Field investigation showed that virus-induced symptoms of tobacco include mosaic, leaf malformation, necrotic rings, vein necrosis, dwarf and etch. The incidence of tobacco virus diseases at nine survey spots is between 28%-84% and the disease index is between 6.67-60.89. The results revealed that no virus was detectable in samples at the seeding stage and PVX was undetectable in samples at the elongating stage. All of the 7 viruses were detected at the late growth stages. Both TMV and CMV had higher detection ratios than those of TuMV and PVY while PVX had the lowest detection ratio. These results showed that tobacco viral diseases commonly occur in Fengdu and TMV and CMV are the prevalent pathogenic viruses.
