    响应面法优化全自动索式提取仪测定烟草石油醚提取物条件

    Optimization of Extraction for the Petroleum Ether Extract from Tobacco with Fully-Automatic Soxhlet Extractor by Response Surface Methodology

    • 摘要: 为确定全自动索式提取仪提取烟草中石油醚提取物的最佳工艺条件,在单因素试验基础上,以烟草标准物质GBW08514a 石油醚提取物质量分数为指标,利用Design-Expert 软件响应面法优化其提取条件。结果表明,最佳提取条件为:煮沸时间/淋洗时间15/90 min、提取温度100 ℃、提取溶剂量85 mL,此条件下,GBW08514a 石油醚提取物质量分数为4.70%(RSD/%=1.85)。利用响应面法优化得到全自动索式提取仪提取烟草中石油醚提取物的最佳工艺条件,该方法与行业标准的测定结果差异较小,且大大缩短工作时间、提高工作效率,适用于烟草石油醚提取物的批量检测。

       

      Abstract: In order to determine the optimal experimental parameters for the extraction of petroleum ether extract from tobacco with fully-automatic Soxhlet extractor, the single factor tests and response surface methodology (RSM) were executed to optimize the extraction parameters. The flue-cured tobacco standard material of petroleum ether extract (GBW08514a) was used. The experimental results showed that the optimal extraction parameters were as follows: boiling time/leaching time: 15/90 min, extraction temperature: 100 ℃, extraction solvent volume: 85 mL. Under the optimal parameters, the content of petroleum ether extract of the flue-cured tobacco standard material was 4.70% (RSD/%=1.85). Consequently the optimum parameters with fully-automatic Soxhlet extractor for the extraction of petroleum ether extract from tobacco were established by response surface methodology. The discrepancy of the results applied with the proposed method and the traditional method was negligible. Moreover this method saved the labor time and greatly improved working efficiency, which was more suitable for the rapid determination of the petroleum ether extract for a multitude of tobacco samples.
