    内生解淀粉芽孢杆菌Xe01的鉴定及其发酵条件优化

    Identification and Fermentation Optimization of Antagonistic Bacillus amyloliquefaciens Xe01

    • 摘要: 为获得对烟草青枯病菌具有拮抗作用的生防菌,从湖南永州青枯病发病较重烟田的健康烟株根系中分离获得1株对烟草青枯病菌具有较强抑菌活性的菌株,对峙培养其抑菌圈直径达到30.5 mm,将其命名为Xe01。利用形态观察、生理生化特征及16S rRNA基因序列分析鉴定其为解淀粉芽孢杆菌(Bacillus amyloliquefaciens),温室盆栽和小区试验中Xe01对烟草青枯病的防治效果分别达到56.32%和61.46%。对其发酵培养基及发酵条件进行单因素试验及正交设计优化,最适培养基碳源、氮源和无机盐种类及浓度为葡萄糖9.0 g/L、牛肉膏3.0 g/L、氯化钙4.0 g/L;最佳发酵条件为pH 7.0,30℃,180 r/min,培养52 h,使用优化后的发酵条件培养,抑菌圈直径由优化前的30.5 mm提高至42.5 mm,提高了39.34%。研究结果为解淀粉芽孢杆菌Xe01生防菌剂的开发及应用提供了理论基础。

       

      Abstract: To obtain antagonistic biocontrol bacteria for tobacco bacterial wilt control, a strain (Xe01) with strong antibacterial activity against Ralstonia solanacearum was isolated from the roots of healthy tobacco plants in Yongzhou, Hunan Province. It was found that the diameter of inhibition zone was 30.5 mm by confrontation culture method, and the strain was identified as Bacillus amyloliquefaciens by morphological observation, physiological and biochemical characteristics and 16S rRNA gene sequence analysis. The control effect of antagonistic bacterium Xe01 on tobacco bacterial wilt was 56.32% in greenhouse and 61.46% in plot test. The optimum medium for the antagonistic strain Xe01 was glucose 9.0 g/L, beef extract 3.0 g/L, calcium chloride 4.0 g/L, and the optimum fermentation conditions were pH 7.0, 30℃, 180 r/min, culture 52 h. Under optimized fermentation conditions, the diameter of inhibition zone increased from 30.5 mm to 42.5 mm, increased by 39.34%. The results from this study provided a theoretical basis for the development and application of biocontrol agent using Bacillus amyloliquefaciens Xe01.
