    干旱胁迫对转BnDREB1-5烟草碳氮代谢和渗透调节物质的影响

    Effects of Drought Stress on Carbon and Nitrogen Metabolism and Osmotic Adjustment Substances of Transgenic Tobacco with BnDREB1-5 Gene

    • 摘要: 在干旱胁迫后检测了转BnDREB1-5基因烟草碳氮代谢指标和渗透调节物质的变化。结果表明,干旱胁迫后转基因烟草碳代谢指标淀粉酶活性、转化酶活性和可溶性糖含量都明显升高,野生型烟草淀粉酶活性和可溶性糖含量稍微升高,转化酶活性反而降低;转基因烟草氮代谢指标硝酸还原酶活性、可溶性蛋白质含量稍微下降,而野生型烟草两者都明显降低;转基因烟叶中有BADH活性,干旱胁迫后BADH活性升高,而野生型烟草中检测不到BADH活性;干旱胁迫后转基因烟叶中脯氨酸含量是野生型的1.08倍。因此,干旱胁迫后转基因烟草碳氮代谢、防御酶活性等方面明显优于野生型。

       

      Abstract: After drought stress the changes of the carbon and nitrogen metabolism and osmotic adjustment on tobacco with 35S: BnDREB1-5 transformation were analyzed. The results indicated that after physical drought-stress the carbon metabolisms of transgenic tobacco, which included the amylase activity, the invertase activity and the soluble sugar content, increased significantly, and the amylase activity and the soluble sugar content of wild-type increased slightly, but the invertase activity of wild-type decreased. The nitrogen metabolisms of transgenic tobacco, which included the nitrate reductase activity and the soluble protein content, decreased slightly, while those of wild-type tobacco decreased significantly. The BADH activity of transgenic tobacco leaves was detectable and increased after drought stress, but that of wild-type was not detectable. After drought stress, the proline content of transgenic tobacco leaves was 1.08 times as large as that of wild-type. After drought stress the carbon and nitrogen metabolism and defense activity on tobacco with 35S: BnDREB1-5 transformation were obviously better than wild-type.
