    烟草花粉活力鉴定方法筛选

    Screening for Optimum Method of Tobacco Pollen Vigor Evaluation

    • 摘要: 以白肋烟B37花粉为试验材料,对花粉活力鉴定方法进行了分析比较,并设计了培养配方、培养温度、培养时间等15种花粉活力鉴定方法。结果表明,花粉离体培养萌发测定花粉活力是较为理想的烟草花粉活力鉴定方法,其中葡萄糖(3%)+硼酸(50 mg/L)+CaCL2(20 mg/L)、蔗糖(10%)+硼酸(50 mg/L)+CaCL2(20 mg/L)25 ℃恒温暗箱保湿培养3 h,花粉萌发率最高,新鲜花粉活力分别为86.8%和91.3%。

       

      Abstract: With the experimental material of Burley B37 pollen, various tobacco pollen vigor evaluation methods were compared. The results indicated that pollen germination was the best method for identifying pollen vigor. With the formula of 3% glucose, 50 mg/L boric acid, 20 mg/L CaCl2 and 10% sucrose, 50 mg/L boric acid, 20 mg/L CaCl2, cultivating 3 h under 25 ℃ constant temperature, the percentage of pollen germination was the highest. The pollen vigor was 86.8% and 91.3% respectively.
