    Cloning and Sequence Analysis of Potassium Ion Channel Gene NKT5 in Nicotiana Tabacum

    • 摘要: 本研究运用已经获得的1 条490 bp 的普通烟草钾离子(K+)通道基因的中间片段来设计特异性引物,应用RACE方法获得其3'和5'末端cDNA 序列。通过三段cDNA 序列拼接结合全长克隆测序验证,获得一个未报道的普通烟草K+通道基因,并将其命名为NKT5(NCBI 登录号为HM010922)。NKT5 的cDNA 全长为2 365 bp,其中5'端非编码区190 bp、3'端非编码区249 bp、编码区1 926 bp,编码641 AA。对该基因进行了序列分析及功能预测,为进一步阐明其在烟草吸钾及抗逆过程中的功能及分子机制奠定了基础。

       

      Abstract: Based on a 490 bp middle segment of K ion channel gene, a pair of specific primers was designed to acquire 5' and 3'-ends of cDNA by RACE strategy. An unreported K ion channel gene in tobacco (Nicotiana tabacum) named as NKT5 was obtained by splicing three cDNA sequences and sequencing full-length cloning. The full-length cDNA of NKT5 is 2 365 bp long with 190 bp 5'-untranslated region (UTR), 249 bp 3'-UTR and 1 926 bp coding sequence. The deduced amino acid is 641 AA long. The gene sequence analysis and function prediction were carried out in order to clarify molecular mechanisms of potassium absorption and stress-resistance in the tobacco.
